事故で突然、父・貴志（橋爪淳）を亡くした女子高生・星泉（長澤まさみ）の前に、浅草の弱小ヤクザ目高組の若頭・佐久間（堤真一）が組員たちを引き連れて現れた。佐久間は、先代組長を亡くして解散寸前の目高組の存続のため、唯一の血縁者である泉に「組長」を襲名してもらえないかと頼み込む。寝耳に水の話に初めは拒み続ける泉だが、結局は目高組八代目組長を襲名してしまう…。そんな折、泉は警察から父の死は他殺の可能性もあると伝えられる…。また、葬儀に父の遺影をじっと見つめる謎の女・真由美（小泉今日子）の姿が…。更には、麻薬に手を出す悪道ヤクザ・浜口組と目高組との対立が激化。そして、表向きは麻薬撲滅を掲げる大物政治家・三大寺（緒形拳）までもが現れ、事態は一筋縄ではいかない展開となっていく…。
